Galantos Pharma develops Nicotinic Allosteric Potentiating Ligands (APLs), using galantamine’s APL activity as a lead. Galantamine is a clinically proven natural compound that acts as both a reversible inhibitor of the enzyme acetylcholinesterase (AChE) and as an APL. To optimize APLs and to adapt the therapeutic principle to a given indication, three properties require modification:

1. Reduction or complete removal of the activity as an AChE inhibitor
2. Optimization of nAChR binding and sensitization for subtype specificity and efficacy,
3. Improvement of ADME properties.

Galantos presently develops four types of optimized APLs, of which only few are derivatives of galantamine:

1. Memogain®, a galantamine pro-drug with improved brain penetration
2. APL without any AChE inhibitory activity,
3. APL with higher potency in sensitizing nAChR, and
4. APL that act selectively on distinct subtypes of nAChR,.

Memogain® is the most advanced product of Galantos Pharma.

Among the other compounds are pharmacophores for the interaction with nicotinic receptors and cholinesterases, and lead structures have been selected on the basis of additional in-vitro models.
